RNA Sequencing on a large Scale

RNA sequencing makes it possible to sequence the transcriptome in a specific tissue at a specific developmental stage [2]. This snapshot of the RNA molecules present can be used to draw conclusions about gene expression and detect post-transcriptional modifications [1]. RNA data represent a unique source for the discovery of new biomarkers, the prediction of diseases and the development of innovative drugs. Therefore, the ability to perform a large number of RNA sequencing experiments quickly and cost-effectively is a concrete need for many researchers. And exactly this need is fulfilled by the products of Alithea Genomics!

BRB-Seq™ simplifies the Generation of large RNA Data

To make large-scale RNA sequencing available to everyone, the founders of Alithea Genomics have developed a new technology called Bulk RNA Barcoding and sequencing, or BRB-seq™ for short [3]. The innovative sequencing method uses optimized, barcoded primers, the so-called BRB-seq™ oligos, which label RNA samples in a unique and sample-specific way (Fig. 1). The use of these oligos enables the generation of consistent, reproducible and uniform RNA sequencing data [4].

BRB-seq™ is based on the optimized BRB-seq™ oligos, which consist of an oligo-dT sequence, a unique molecular identifier (UMI), the sample barcode and an adapter to the sequencing interface. In the first step of library preparation, the BRB-seq™ oligos bind to the 3'-end of an RNA molecule to be sequenced, thereby giving it a sample-specific label [4].

Since RNA barcoding is at the beginning of library preparation, samples can be pooled and processed together in subsequent steps. Using Alithea Genomics' technology, up to 384 samples can be analysed together in one reaction tube. Such massive sample multiplexing leads to a drastic reduction in reagent and plastic consumption as well as manual work steps, which is why the hands-on time for the entire workflow is around 2.5 hours [4]. 

In addition, the technology is compatible with numerous sample types: from purified RNA (BRB-seq) to cells and organoid lysates (DRUG-seq), Alithea offers solutions for a wide range of starting material. In addition, the kits can be used with Illumina™ sequencers as well as with MGI devices. The Swiss company's first-class sequencing kits make large-scale RNA data generation accessible to everyone.
