IDH1, cytoplasmic enzyme, belongs to the isocitrate and isopropylmalate dehydrogenases family. This protein catalyzes the third step of the citric acid cycle, which involves the oxidative decarboxylation of isocitrate, forming alpha-ketoglutarate and CO2 in a two step reaction. The first step involves the oxidation of isocitrate to the intermediate oxalosuccinate, while the second step involves the production of alpha-ketoglutarate. One unit will oxidize 1.0pmole of DL-Isocitrate to D-alpha-Ketoglutarate per minute in the presence of beta-NADP at pH7.4 at 37°C. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
