Fatty Acid Synthase (FASN) catalyzes the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FASN is fused in-frame with the Cterminus of ER-alpha. FASN is highly expressed in adipocytes and liver. FASN is a central enzyme in de novo lipogenesis. FASN is a target for SREBP and is upregulated by LXR activation, it is also one of the accepted markers for insulin resistance, SREBP and LXR activation. FASN: Bovine: 2513 aa, Pig: 2411 aa, Chick: 2511 aa, human: 2511 aa, Rat: 2505 aa, mouse: 2504 aa, 272.4kD, Chromosome: 11E2. Expression: high in brain, lungs, and liver. Source: Synthetic peptide, 18-aa corresponding to Mouse FASN (KLH). , Applications: Suitable for use in ELISA and Antibody Blocking. Other applications not tested. Recommended Dilution: ELISA: 50-100ng., Antibody Blocking: 5-10ug /1ug of Affinity Purified antibody or 1ul of serum., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer or ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
