Factorxis the only known physiological activator of thrombin and plays a critical role in blood coagulation. Factorxis also involved in cell surface responses including cell activation, gene expression and mitogenesis. Factorxis a serine protease that is synthesized in the liver and released into the bloodstream. Mouse Factorxconsists of a single-chain precursor of 481aa residues that contains a signal peptide (residues 1-20), pro region (residues 21-40), light chain (residues 41-180) and heavy chain (residues 184-481). The light and heavy chains are linked by a disulfide bond. The light chain contains a Gla and two EGF-like domains and the heavy chain corresponds to the serine protease domain. The full length mouse Factorxwas expressed and purified. Upon activation, the active protease was further purified and analyzed for activity against a peptide substrate. In addition to the activity described in the Activity Assay Protocol, Factorxcan also be used to cleave fusion proteins containing a Factorxcleavage site. The conditions for the optimal cleavage of a particular fusion protein must be determined for each individual fusion protein. Source: Recombinant corresponding to aa1-481 from Mouse Coagulation Factor X, CF fused with 10-His-tag at C-terminal expressed in Sf21 cells. Molecular Weight: ~29kD, ~10kD, Endotoxin: <1EU/1ug (LAL) , Biological Activity:, Measured by its ability to cleave the fluorogenic peptide substrate, Mca-RPKPVE-Nval-WRK(Dnp)-NH2. The specific activity is >300 pmol/min/ug, as measured under the described conditions. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
