Rottlerin (Mallotoxin)

Rottlerin is an inhibitor of PRAK (IC50 1.9M), MAPKAP Kinase 2 (IC50 5.4M) and CaM Kinase III (3-6M). Rottlerin inhibits PMA-induced edema in mouse ear. For use as an Inhibitor of PRAK, MAPKAP kinase 2, and CaM Kinase III activity. Also shown to act as a direct mitochondrial uncoupler. Thought to stimulate autophagy by targeting upstream mTORC1 control pathways. Appearance: Supplied as an orange-brown powder. Purity: One spot, Rf=0.85, Melting Point: 200-205°C(lit.), Solubility: Soluble in DMSO, chloroform or 100% ethanol. Not soluble in water. Handling: Warning! Toxic. Wear gloves/mask when handling product. Avoid contact by all modes of exposure. Storage and Stability: Powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with DMSO, chloroform, or 100% ethanol. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
