Nocodazole, an inhibitor of microtubule formation, was found to greatly inhibit cell division around the mound stage as well as during the vegetative growth phase, when applied to exponentially growing Ax-2 cells. Mitotic arrest of cells with nocodazole can be used to study phosphorylation of microtubule-associated proteins which increase during mitosis, concomitant with increased microtubule dynamics. Nocodazole has been found to dramatically inhibit T cell antigen receptor (TCR) signaling, independently of its ability to depolymerize microtubules. Nocodazole arrests the cell cycle at G2/M phase and prevents phosphorylation of the T cell antigen receptor and inhibits its activity. Due to insolubility (nocodazole is only slightly soluble in water), no more than 10ug/ml concentration can be used in cell culture systems. Nocodazole is stable in biological media for seven days. Storage and Stability: May be stored at RT for short-term only. Long-term storage is recommended at -20°C for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
