JC2-11 is an inflammasome inhibitor. It inhibits increases in IL-1beta production induced by the NOD-like receptor protein 3 (NLRP3) activators ATP (Cay-14498, Cay-40182), nigericin (Cay-11437), or monosodium urate crystals in LPS-primed primary mouse bone marrow-derived macrophages (BMDMs) when used at concentrations of 50 or 100 µM. JC2-11 (100 µM) inhibits increases in IL-1beta production induced by flagellin or dsDNA, which are activators of NLR family CARD domain-containing protein 4 (NLRC4) and absent in melanoma 2 (AIM2), respectively, in LPS-primed primary mouse BMDMs. It also scavenges DPPH (Cay-14805) radicals in a cell-free assay (IC50 = 52.5 µM) and inhibits LPS-induced nitric oxide (NO) production in primary rat microglial cells.
