A-867744 is a positive allosteric modulator of alpha7 nicotinic acetylcholine receptors (nAChRs). It is selective for alpha7 nAChRs over alpha3beta2- or alpha4beta2 subunit-containing nAChRs (IC50s = 0.023, 19.5 and 6.4 µM, respectively) and the serotonin (5-HT) receptor subtype 5-HT3A (Ki = >30 µM). A-867744 potentiates ACh-induced currents in Xenopus oocytes expressing human alpha7 nAChRs (EC50 = ~1 µM).
