Nadifloxacin is a fluoroquinolone antibiotic. It is active against clinical isolates of S. aureus, coagulase-negative staphylococci, P. acnes, and P. granulosum (MIC50s = 0.03, 0.06, 0.25, and 0.125 mg/L, respectively). Nadifloxacin inhibits the activities of DNA gyrase and topoisomerase IV (IC50s = 8.83 and 3.22 mg/L, respectively, for the S. aureus enzymes). Nadifloxacin (30 µg/ml) inhibits P. acnes-induced increases in the production of IFN-gamma, IL-1beta, and IL-12p70 in human peripheral blood mononuclear cells (PBMCs), as well as reduces P. acnes-induced increases in the production of IL-8 and IL-6 in isolated human keratinocytes. Topical application of a cream containing nadifloxacin (1%) reduces the number of skin lesion colony forming units (CFUs) in a mouse model of burn wounds infected with S. aureus or P. acnes.
