This gene encodes an enzyme involved in catalyzing the conversion of angiotensin I into a physiologically active peptide angiotensin II. Angiotensin II is a potent vasopressor and aldosterone-stimulating peptide that controls blood pressure and fluid-electrolyte balance. This enzyme plays a key role in the renin-angiotensin system. Many studies have associated the presence or absence of a 287 bp Alu repeat element in this gene with the levels of circulating enzyme or cardiovascular pathophysiologies. Two most abundant alternatively spliced variants of this gene encode two isozymes - the somatic form and the testicular form that are equally active. Multiple additional alternatively spliced variants have been identified but their full length nature has not been determined. Sample Type: Serum, plasma, tissue homogenates, Intended Use: For the quantitative determination of mouse angiotensin converting enzyme (ACE) concentrations in serum, plasma, tissue homogenates. Sensitivity: 0.039ng/ml, Range: 1.56-100ng/ml, Specificity: This assay has high sensitivity and excellent specificity for detection of mouse ACE. No significant cross-reactivity or interference between mouse ACE and analogues was observed. Test Principle: This BioAssay(TM) employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ACE has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ACE present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACE is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACE bound in the initial step. The color development is stopped and the intensity of the color is measured. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
