Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine aa's and the phosphorylation of serine or threonine. High levels of phosphorylation of Histone H3 are associated with mitosis. Consistent with this role, Histone H3 phosphorylated at threonine 3 has been shown to be recognized by the BIR domain Survivin, a member of the inhibitor of apoptosis (IAP) family. Applications: Suitable for use in Western Blot and Cell Function Assay. Other applications not tested. Recommended Dilution: Western Blot: 1:8000, Optimal dilutions to be determined by the researcher. Positive Control: Colcemid treated HeLa acid extract and recombinant Histone H3 negative control. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
