Product information "Anti-Tubulin, beta 3 (Tubulin beta III, Tubulin beta-3 Chain, TUBB 3, TUBB3, Neuron Specific beta II"
The cytoskeleton consists of three types of cytosolic fibers: microtubules, microfilaments (actin filaments), and intermediate filaments. Globular tubulin subunits comprise the microtubule building block, with a/b-tubulin heterodimers forming the tubulin subunit common to all eukaryotic cells. y-tubulin is necessary to nucleate polymerization of tubulin subunits to form microtubule polymers. Many cell movements are mediated by microtubule action, including the beating of cilia and flagella, cytoplasmic transport of membrane vesicles, chromosome alignment during meiosis/mitosis, and nerve-cell exon migration. These movements result from competitive microtubule polymerization and depolymerization or through the actions of microtubule motor proteins. beta3-tubulin (TUBB3) is one of six beta-tubulin isoforms and is expressed highly during fetal and postnatal development (axon guidance and maturation). Its expression levels decrease in the adult central nervous system (CNS) but remain high in the peripheral nervous system (PNS). Microtubules enriched in beta3-tubulin are more dynamic than those composed of other beta-tubulin isoforms. Research studies have shown that mutations in the beta3-tubulin gene TUBB3 cause ocular motility defects and other nervous system disorders. Furthermore, beta3-tubulin is present in neoplastic but not in normal differentiated glial cells. Thus, beta3-tubulin is a great neuronal marker. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000 , Immunohistochemistry (Paraffin): 1:50. Use citrate as unmasking buffer., Immunoprecipitation: 1:50, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
