The mammalian SUMO-specific protease (SENP1) is active against sentrin, but not ubiquitin or NEDD8-modified proteins in vivo (1). SENP1 provides a unique tool to study the role of sentrinization in the biological function of Pml and in the pathogenesis of acute promyelocytic leukemia (1). SENP1 localizes to the nucleus mainly in discrete subdomains, a subset of which colocalized with Pml nuclear bodies (NBs) (2). Both The herpes simplex virus 1 (HSV-1) immediate early regulatory protein (ICP0) and SENP1 protease promote the loss of SUMO-1 from the nucleus (2). SENP1 recruitment may be an important (though not sufficient) role in the modulation of SUMO-1 and NBs by ICP0 (2). SENP1 itself may be a target for SUMO-1 modification occurring at a nonconsensus site. SENP1 localization is influenced by expression and localization of SUMO-1-conjugated target proteins within the cell (3). Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Western Blot: 1-3ug/ml, Immunoprecipitation: 7ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
