This gene encodes pyruvate carboxylase, which requires biotin and ATP to catalyse the carboxylation of pyruvate to oxaloacetate. The active enzyme is a homotetramer arranged in a tetrahedron which is located exclusively in the mitochondrial matrix. Pyruvate carboxylase is involved in gluconeogenesis, lipogenesis, insulin secretion and synthesis of the neurotransmitter glutamate. Mutations in this gene have been associated with pyruvate carboxylase deficiency. Alternatively spliced transcript variants with different 5' UTRs, but encoding the same protein, have been found for this gene. Applications: Suitable for use in Peptide ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Peptide ELISA: 1:16,000, Western Blot: 0.03-0.1ug/ml, Optimal dilutions to be determined by the researcher. Positive Conrol: Mouse Liver lysates, Control Peptide: P9543-95A-P: Pyruvate Carboxylase (PC, PCB, Pyruvate carboxylase, mitochondrial, Pyruvic carboxylase) (Control Peptide), Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
