Cyclic GMP-dependent protein kinases (PKGs or cGKs) are classified into two types, PKGI and PKGII based on their historical order of characterization. There are two PKGI isoforms, PKGIalpha (76kD) and PKGIbeta (78kD) which arise from the alternative splicing of a single gene. These two isoforms differ in their amino-terminal autoinhibitory domains but are similar in their cGMP-binding sites and catalytic domains. PKGIalpha is highly expressed in the Purkinje cell of the cerebellum, in platelets, lung and in smooth muscle cells whereas the PKGIbeta isoform is primarily expressed in smooth muscle cells. Studies have shown that cGKs and cyclic AMP-dependent kinases (cAKs) are highly homologous protein kinase families with similar substrate specificities and phosphorylation of cellular proteins by both families of kinases leads to alterations in calcium mobilization, protein phosphatase activity, ion channel function, gene transcription, smooth muscle contractility and platelet aggregation. However, in most of these studies a clear dissection between the cGMP kinase pathway and other pathways such as the cAMP kinase pathway have been difficult to achieve. Recent studies using mice deficient in PKGs have shown that cGMP kinases regulate very specifically distinct pathways which are separate from those used by cyclic AMP kinases. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot (Colorimetric): 1:1000, Immunohistochemistry: Paraffin sections, Optimal dilution determined by the researcher. Positive Controls: Mouse Brain Tissue Extract, Rat Brain Tissue Extract, Human Prostate Tissue, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
