Phospholipases A2 (PLA2s) hydrolyze the sn-2 fatty acid acyl ester bond of phosphoglycerides, releasing free fatty acids and lysophospholipids. They play an important role in a variety of cellular processes, including digestion and metabolism of phospholipids, as well as production of precursors for inflammatory reactions. Phospholipases A2 belongs to PLA2 group II, which consists of extracellular enzymes with low molecular masses that require calcium ions for catalysis. Applications: Suitable for use in Direct ELISA, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilutions: Direct ELISA: 0.5-1.0ug/ml. The detection limit for recombinant human PLA2G2A is ~1.5ng/well., Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant human PLA2G2A is ~2ng/lane under non-reducing conditions and ~50ng/lane under reducing conditions., Immunoprecipitation: Used to immunoprecipitate recombinant human PLA2G2A from conditioned media of transfected Sf21 cells., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
