Product information "Anti-PDK-1 (Pyruvate Dehydrogenase Kinase-1, PDHK1)"
PDPK-1 (3-phosphoinositide dependent protein kinase, gene name PDPK1) is a 58 68kD, 55 aa monomeric protein of the AGC serine/threonine kinase family. It is activated by phosphorylation in the presence of PtdIns(3,4,5) P3 or PtdIns(3,4) P2. Akt, S6 kinases, PKA and PKCZ are reported PDK1 substrates. Through Akt, PDK1 mediates many of the intracellular actions of insulin. Within the region used as an immunogen, human PDK1 shares 98% aa identity with mouse and rat PDK1. One reported isoform has an alternate start site at aa 50, while another lacking aa238263 is predicted to be catalytically inactive. Applications: Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1ug/ml Lysates of Jurkat, human acute T cell leukemia cell line, HepG2 human, hepatocellular carcinoma cell line, Daudi human, Burkitt's lymphoma cell line, A20 mouse B cell, lymphoma cell line, and C6 rat glioma cell line, Immunocytochemistry: 5-25 ug/ml Immersion fixed HeLa human cervical carcinoma cell line, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
