NQO1 (NADPH Quinone acceptor Oxidoreductase 1, also DTD and Menadione reductase) is a 3033kD cytoplasmic member of the NAD(P)H dehydrogenase family of enzymes. It is widely expressed, being found in adipocytes, endothelial cells and hepatocytes. NQO1 serves as a cytoprotective molecule, reducing quinones (atmospheric benzene and esrtrogen compounds) and nitroaromatics without consuming sulfhydryl compounds. It also acts as a backup superoxide scavenger to SOD. NQO1 reportedly binds and stabilizes key cell protection molecules such as p53, thus acting as a gatekeeper for proteosomemediated protein turnover. Human NQO1 is 274 amino acids (aa) in length. It contains an FMN reductase domain (aa5212) and two utilized Lys acetylation sites. NQO1 functions as a noncovalent homodimer. There are three potential at Met22, while two others show a deletion of aa141174 and 102139, respectively. Full length human NQO1 shares 86% aa sequence identity with mouse NQO1. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
