Notch-1 (so named for "notches" in fly wings, also TAN-1) is a 300 kD, 2538aa member of the notch family of type I TM glycoproteins. It is associated with gene activation in both embryo and adult. It undergoes Golgi processing to generate a heterodimer composed of a 180-200kD disulfide-linked extracellular domain (aa18-1664), and a 120kD membrane-bound segment (aa 1665-2556) that is cleaved to form the notch intracellular domain transcription factor upon ligand binding. Human Notch-1 ECD aa19-526, including the first 13 EGF repeats, shows 91% aa identity with corresponding regions of mouse and rat Notch-1, and 60% aa identity with , human Notch-2 and Notch-3. Applications: Suitable for use in ELISA and Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 25ug/10e6 cells using U2OS human osteosarcoma cell line. , ELISA (Capture): 2-8ug/ml, ELISA (Detection): 0.5-2ug/ml used with N5375-03Z as the capture antibody, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
