Product information "Anti-Mast Cell Chymase (CMA1)"
Chymase-1 (also mast cell protease I) is a secreted, 30kD glycoprotein member of the granzyme subfamily, peptidase S1 family of enzymes. It is secreted by mast cells and vascular smooth muscle cells, and converts angiotensin I into angiotensin II. This promotes fibroblast proliferation and secretion of ECM. Mature human chymase is 226aa in length. It contains one peptidase S1 domain (aa 2-245). There is one potential alternate splice form. It shows a deletion of the 99 C-terminal amino acids (aa149-247). Applications: Suitable for use in Direct ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Direct ELISA: 0.5-1ug/ml. Detection limit for rhChymase is ~1ng/well., Western Blot: 1-2ug/ml. Detection limit for rhChymase is , ~10ng/lane under non-reducing and reducing conditions. Chemiluminescent detection wil increase sensitivity by 5 to 50 fold., Immunoprecipitation: 25ug/ml from conditioned cell culture medium., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
