HSP47, also known as heat shock protein 47, colligin, or gp46, is a collagen-specific stress glycoprotein which resides in the ER and has been shown to be involved in the synthesis/assembly of various collagens as a molecular chaperone. The protein belongs to the serpin (serine protease inhibitor) superfamily but unlike other serpin family proteins, it localizes within the endoplasmic reticulum (ER) because of the presence of RDEL (Arg-Asp-Glu-Leu) sequence, a variant of KDEL sequence, at the C-terminus. HSP47 has been shown to bind to the KDEL receptor in the ER. HSP47 is susceptible to phosphorylation. Both gp46 and HSP47 were later found to be heat-inducible and susceptible to phosphorylation. It is generally believed that HSP47 binds procollagen immediately after it co-translationally enters the ER, and it dissociates from procollagen before its secretion. When cells are heat shocked or treated with inhibitors of the triple-helix formation of procollagen, HSP47 is retained to these abnormal or denatured procollagens for much longer periods of time. Expression of HSP47 on cell surface following alkalization of the endosomal compartments under conditions of stress suggest HSP47 may be a serpin family protein that may modulate cell migration during development and invasion and metastasis in cancer. HSP47 has been suggested to play an important role in tumor metastasis and can be used as a prognostic marker for the metastatic activity of human tumor cells. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
