The activity of Glutamate/Aspartate Transporter 2 (GLT1, EAAT2, Excitatory Amino Acid Transporter 2), the main glutamate transporter in the CNS, can be regulated by protein kinase C (PKC). It is known that activation of PKC by phorbol esters promotes the clathrin-dependent internalization of the transporter, followed by its lysosomal degradation. However, the molecular mechanisms that link PKC activation and the internalization of the Glutamate/Aspartate Transporter 2 are not fully understood. The internalization process may play a role in the ubiquitylation of lysine residues located in the C-terminal tail of the Glutamate/Aspartate Transporter 2. Exposure to PMA increases the ubiquitylation of the Glutamate/Aspartate Transporter 2 in transfected cells and in the rat brain, and this ubiquitylated Glutamate/Aspartate Transporter 2 accumulates in the intracellular compartment. The activation of PKC induces the ubiquitylation of these C-terminal lysine residues in the Glutamate/Aspartate Transporter 2 and this modification mediates the interaction of the transporter with the endocytic machinery. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.5ug/ml detected Glutamate/Aspartate Transporter 2 on 10ug of mouse brain tissue lysate., Optimal dilutions to be determined by the researcher. Positive Control: Mouse brain tissue lysate, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
