Death receptor 4 (DR4) or TRAIL-R1, a member of the tumor necrosis factor receptor superfamily, is a cell surface receptor that triggers the apoptotic machinery upon binding to its ligand tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). DR4 is expressed in most of human tissues including spleen, peripheral blood leukocytes, small intestine and thymus. DR4 expression is enhanced by DNA damage whether induced by ionizing radiation or by chemotherapeutic agents. On binding DR4, TRAIL induces cell death via caspase-dependent apoptosis. DR4 mediate oligomeric Abeta induction of extrinsic apoptotic pathways in human microvascular cerebral endothelial cells with activation of both caspase-8 and caspase-9. It is considered a promising molecular target for cancer therapy. Protein function: Receptor for the cytotoxic ligand TNFSF10/TRAIL (PubMed:26457518). The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis.
