Duffy Antigen Receptor for Chemokines (DARC), also known as Fy glycoprotein and CD234, is a 7TM non-signaling chemokine receptor that is expressed on venular endothelial cells and erythrocytes. DARC binds and mediates the internalization of several inflammatory CC and CXC chemokines. It transports chemokines to the lumenal face of venular endothelium to promote the extravasation of leukocytes to sites of inflammation. DARC polymorphisms are important in blood transfusion compatibility and determine the susceptibility of erythrocytes to Plasmodium vivax infection. Human DARC shares 60% amino acid sequence identity with mouse DARC. Applications: Suitable for use in Flow Cytometry and Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 25ug per 10e6 human blood erythrocytes, Immunocytochemistry: 8-25ug/ml using immersion fixed HUVECs, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
