Cyclin D1 gene is identical with the BCL-1 and the PRAD-1 oncogene. It is part of the chromosome 11q13 amplicon which is amplified in several common tumor types like breast, lung, esophagus, urinary bladder and head and neck carcinoma. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. About 50-70% of mantle cell lymphomas , (MCL) contain a t (11, 14)(q13, q32) translocation resulting in over-expression of cyclin D1. Cyclins were originally identified as proteins whose levels fluctuated during the cell cycle. Subsequent studies have shown the cyclins control entry into different phases of the cycle cell by regulating the activity of cyclin-dependent kinases (CDKs). Mitotic or G2 cyclins regulate entry into M phase. G1 cyclins regulate entry into S phase. Cyclin D1, a G1 cyclin, is essential for control at the G1/S transition (START in yeast). It regulates the activity of CDK4 and CDK6 protein kinases. Translocation of the gene encoding cyclin D1, the bcl-1 oncogene, is associated with human B-lymphocytic malignancy, suggesting that alteration of cell cycle progression may be oncogenic. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: ELISA: 1:500-1:2000, Western Blot: 1:500-1:1000, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
