Acetyl-CoA carboxylase (ACC) catalyzes the pivotal step of the fatty acid synthesis pathway. The 265kD ACC alpha is the predominant isoform in liver, adipocytes and mammary gland, while the 280kD ACC beta is the major isoform in skeletal muscle and heart (1). Phosphorylation by AMPK at Ser79 or by PKA at Ser1200 inhibits the enzymatic activity of ACC (2). ACC is a potential target of anti-obesity drugs (3,4). Applications: Suitable for use in ELISA, Immunofluorescence, Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000, Immunohistochemistry (Paraffin): 1:100 Citrate/TBST , Immunofluorescence (IF-IC): 1:200, Flow Cytometry: 1:400, Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb, I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb, I1904-46Q: IgG, H&L (HRP) Pab Gt xRb, Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
