Anti-Granzyme B

Granzyme B, also known as CTLA-1, is a serine protease found in the cytotoxic granules of cytotoxic T lymphocytes and NK cells and in articular chondrocytes. Granzyme B enters target cells through perforin channels and induces caspase-dependent apoptosis. It is upregulated in rheumatoid arthritis and cleaves vitronectin, fibronectin, and laminin. Human Granzyme B is synthesized as a precursor with a signal peptide (aa 1-18), a two amino acid propeptide, and a mature chain (aa 21-247). Mature human Granzyme B shares 69% amino acid sequence identity with mouse Granzyme B. Applications: Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry, Direct ELISA. Other applications not tested. Recommended Dilution: Western Blot: 0.5ug/ml NK-92 human natural killer lymphoma cell line., Flow Cytometry: 2.5ug/10^6 NK-92 human natural killer lymphoma cell line fixed with paraformaldehyde and permeabilized with saponin. , Immunocytochemistry: 8-25ug/mL Immersion fixed human peripheral blood mononuclear cells., Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
