Glutamate is a major excitatory neurotransmitter. One enzyme central to the metabolism of glutamate is glutamate dehydrogenase GDH1, that catalyzes the reversible deamination of L-glutamate to 2-oxoglutarate using NAD+ or NADP+. Mammalian GDH is composed of six identical subunits, and the regulation of GDH is very complex. It has been a major goal to identify the substrate and regulatory binding sites of GDH. It is only in recent years that the three-dimensional structure of GDH from microorganisms is available. Very recently, crystallization of bovine liver GDH was reported for the first time from the mammalian sources. However, remarkably little is known about the detailed structure of mammalian GDH, especially the brain enzymes. Bovine glutamate dehydrogenase exists as a homohexamer located within the mitochondrial matrix. Applications: Suitavle for use in ELISA, Western Blot, Dot Blot, Immunoperoxidase Electron Microscopy and Immunohistochemistry. Other applications have not been tested. Recommended Dilution: ELISA: 1:1000-1:4000, Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Labeled with horseradish peroxidase (HRP). Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
