One of the many phosphodiesterases that compartmentalize and hydrolyze cAMP and cGMP into AMP and GMP is PDE10. Only one member of the cAMP and cGMP-specific PDE10 family has been cloned and characterized. The PDE10A gene is expressed in at least two variant forms (PDE10A1, PDE10A2) generated by RNA splicing and/or the use of alternate initiation sites. PDE10A is widely expressed in the putamen and caudate nucleus in the brain, thyroid and testis. The expression of PDE10A is also abundant in Heart. Molecular Weight: 114.5kD , Specific Activity:, 599pmol/min/ug, Assay Conditions: 10mM Tris-HCl, pH7.4, 10mM MgCl2, 1mM MnCl2, 200uM cAMP, 2.5kU 5' nucleotidase, 37ºC, 20min. Application: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Recommended Dilution: Optimal dilution determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
