CD40 belongs to the TNF-receptor superfamily and is essential in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development and germinal center formation. The interaction of CD40-CD40L is necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. CD40 is constitutively expressed by antigen presenting cells, including dendritic cells, B cells and macrophages. Consistent with its widespread expression on normal cells, CD40 is also expressed on a wide range of tumor cells, including non-Hodgkin's and Hodgkin's lymphomas, myeloma and some carcinomas including nasopharynx, bladder, cervix, kidney and ovary. Sequence: The Fc portion of human IgG1 is fused to the C-terminus of human CD40 (aa 21-193), expressed in HEK 293 cells., Binds human and mouse CD40L. Biological Activity: Blocks the binding of CD40L to CD40. Molecular Weight: ~54kD (SDS-PAGE), Endotoxin: <0.01EU/ug (LAL test, Lonza). Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
