Thrombopoietin receptor (Tpo R), also known as myeloproliferative leukemia protein (c-mpl), is a 95kD type I transmembrane protein that is a member of the type I cytokine receptor family within the hematopoietin/cytokine receptor superfamily. The 635aa full-length human Tpo R contains a 25aa signal sequence, a 466aa extracellular domain with a ligand binding domain and two fibronectin type III domains, a transmembrane (TM) domain and a cytoplasmic domain. The extracellular domain of human Tpo R shares 78%, 76%, 81%, 82% and 80% aa identity with mouse, rat, cow, dog, and horse, respectively. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 cells using TF-1 human erythroleukemic cell line., Optimal dilutions to be determined by the researcher. Flow Cytometry: 1. TF-1 cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction., 2. After blocking, 10ul of T5050-50B was added to up to 1x10e6 cells and incubated for 30 minutes at RT. , 3. Unbound antibody was removed by washing the cells twice in F4568-10: Flow Cytometry Staining Buffer., Note: whole blood requires a RBC lysis step at this point., 4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with PE-labeled rmouse IgG2a antibody. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
