Anti-Notch1 (Notch Homolog Protein 1, hN1, Translocation-associated Notch Protein TAN-1)

Notch is synthesized in the endoplasmic reticulum as an inactive form which is proteolytically cleaved by a furin-like convertase (S1 cleavage) in the trans-golgi network before it reaches the plasma membrane to yield an active, ligand-accessible form. Cleavage results in a C-terminal fragment N(TM) and a N-terminal fragment N(EC). Following ligand binding, it is cleaved (S2 cleavage) by TNF-alpha converting enzyme (TACE) to yield a membrane-associated intermediate fragment called Notch extracellular truncation (NEXT). This fragment is then cleaved by presenilin-dependent gamma-secretase (S3 cleavage) to release the intracellular domain (NICD) from the membrane. Applications: Suitable for use in, ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: ELISA: 1:20,000-1:100,000 , Western Blot: 1:1000-1:10,000, Immunohistochemistry: 1:500-1:2500, Optimal dilutions to be determined by the researcher. *Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb, I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb, I1904-46Q: IgG, H&L (HRP) Pab Gt xRb, Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
