Produktinformationen "Anti-Glucosylceramidase (GBA, Alglucerase, GBA1, Imiglucerase)"
Glucosylceramidase is a lysosomal enzyme that cleaves the betaglucosidic linkage of glucosylceramide, an intermediate in glycolipid metabolism. The mature enzyme has 497 amino acids with a molecular weight of 62kD. Glycosylation occurs at four of five Nglycosylation sites and is essential for the trafficking and activity of the enzyme. The enzyme is activated in lysosomes by saposin C, although the mechanism of activation is not well understood. Defects in Glucosylceramidase are the cause of Gaucher disease, also known as glucocerebrosidase deficiency. Gaucher disease is the most prevalent lysosomal storage disease, characterized by accumulation of glucosylceramide in the reticuloendothelial system. Symptoms of Gaucher disease may include enlarged spleen and liver, liver malfunction, skeletal disorders and bone lesions, severe neurologic complications, swelling of lymph nodes, anemia, low blood platelets and yellow fatty deposits on the white of the eye. Currently, enzyme replacement therapy is used to treat patients with the disease. Applications: Suitable for use in ELISA, Western Blot, Simple Western, and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot (LNCaP): 1ug/ml, Simple Western (LNCaP, HEK293T): 10ug/ml, Immunocytochemistry (HeLa): 825ug/ml, Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
