BMPR-IB, also known as ALK-6, is a type I serine/threonine kinase receptor that associates with a type II TGF-b receptor superfamily member to form a heteromeric receptor signaling complex. BMPR-IB is expressed during embryogenesis and in adult brain, bone, and hematopoietic stem cells. Applications: Suitable for use in ELISA and Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 cells using PC-3 human prostate cancer cell line and human iPS cells differentiated to mesoderm., Optimal dilutions to be determined by the researcher. Hybridoma: Myeloma with B cells from Balb/c mice. Flow Cytometry Protocol: This antibody has been tested for Flow Cytometry using PC3 cells., 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction., 2. After blocking, 10ul of B2553-63F was added to up to 1x10e6 cells and incubated for 30 minutes at RT., 3. Unbound antibody was removed by washing the cells twice in Flow Cytometry staining buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry human lyse buffer., 4. The cells were resuspended in Flow Cytometry staining buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG2b antibody. This procedure may need to be modified depending upon the cell type and final utilization. Individual users may need to titrate to determine the optimal reagent amount for their specific use. Storage and Stability: Store at in the dark 4°C. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Caution: APC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
